Excitation source, wavelength, emission filtering, camera response, exposure, enclosure, and operator safety must be planned as one system. Fix geometry and exposure when samples will be compared, and capture a visible-light reference. Fluorescence intensity is not automatically a calibrated chemical measurement, so quantitative work needs an appropriate standard, procedure, and validation beyond a visually successful image.
