What prevents an image scale from becoming inaccurate during capture, stacking, cropping, or resizing?

This staff-authored guidance is generalized. Project-specific and private details should be handled through direct support.

Calibrate with a suitable stage micrometer or traceable reference at the same optical configuration, camera resolution, and processing dimensions used for the sample. Record the pixel scale and do not reuse it after magnification, adapters, focus geometry, crop, or resize changes without recalculation. For stitched images, verify the completed mosaic and preserve the calibration method with the delivered file.
